Cytokines

Cytokines are a class of small molecule proteins or polypeptides that are stimulated, synthesized or secreted by immune cells (such as monocytes, macrophages, T cells, B cells, NK cells, etc.) and some non-immune cells (endothelial cells, epidermal cells, fibers mother cells, etc.).  Cytokines can transmit information between cells.  Cytokines have immune regulation and effector functions including the regulation of innate immunity, adaptive immunity, hematopoiesis, repair of damaged tissues, cell growth, and differentiation. Cytokines play an important role in the immune system by regulating the intensity and duration of immune responses.

Immune cell therapy, represented by CAR-T/NK cells has shown significant therapeutic effects in the treatment of various malignant tumors such as leukemia, lymphoma, and multiple myeloma. As more and more immune cell therapies enter the stage of clinical research, quality management systems have attracted more attention from the industry. During the process of immune cell therapy products, cytokines such as IL-15, IL-7, IL-21 are used as a key raw material for T/NK cell activation and amplification. FDA and Chinese Pharmacopoeia have relevant regulations about the use of these key materials. The FDA CMC recommends using FDA-approved or clinical-grade materials. For Chinese Pharmacopoeia regulations, priority should be given to the use of low-risk materials such as GMP grade materials as opposed to non-GMP grade materials. Therefore, safe, effective, and compliant cytokines are crucial for the success of R&D processes and applications of immune cell therapy drugs.

Anti-idiotypic Antibodies

An idiotope is the unique set of antigenic determinants (epitopes) of the variable portion of an antibody. An anti-idiotypic (Anti-ID) antibody binds to the idiotope of another antibody, usually an antibody drug, which makes it a very powerful tool for antibody drug development, especially for immunogenicity and PK/PD analysis.

To support preclinical/clinical immunogenicity and PK analysis, ACROBiosystems has developed a series of high-affinity anti-idiotypic antibodies. Our pipeline covers five hot targets including adalim*mab, Ritux*mab, Cetux*mab, Trastuz*mab, and Bevaciz*mab. ELISA Kits

Pharmacokinetics (PK) is a branch of science dedicated to the quantitative analysis of absorption, distribution, metabolism and excretion of drug molecules within the body of a living organism. All pre-clinical and clinical studies include the measurement of serum drug concentration, both in animals and in patients, at different points after drug administration. The result is an important indicator of the drug’s pharmacokinetic properties, and is pertinently relevant to dosing recommendations.

The booming market of biologic drugs, driven by a flurry of success with monoclonal antibodies, brings the need for standard high throughput assays to evaluate the content of mAbs in serum samples. Traditionally, the analyses are conducted by indirect ELISA, or with the help of anti-drug antibodies (ADA), each with its own pros and cons. We have developed a series of assays based on the competitive ELISA method. The assay employs a biotinylated mAb against the same target as a tracer to compete with the unlabeled analyte, and uses the interaction between biotin and streptavidin for the eventual readout. This method does not rely on ADAs, and it also alleviates the background issue often occurred with the traditional assay format.

We have launched kits for the studies of CTLA-4, PD-1, and HER-2, respectively, in both humans, and common experimental animals.
