Atlas Plex Workflow: Iterative, Multiplex Detection
AtlasPlex enables iterative multiplexed immunohistochemistry for 3 or 5 protein detection in fixed tissue.
In the workflow, biotinylated primary antibodies (Ab1,...Ab5) are sequentially pre-complexed with StreptaClick®-HRP, allowing for precise, enzyme-mediated tyramide signal amplification.
Each cycle follows the same sequence:
(1) Antibody Complexing: The selected biotinylated antibody is bound to StreptaClick®-HRP.
(2) Tissue Incubation: The Ab-HRP complex is applied to fixed tissue for specific antigen binding.
(3) Tyramide Amplification: HRP catalyzes fluorescent signal deposition at the antigen site.
(4) HRP Quenching: The enzyme is inactivated to prevent cross-reactivity in the next round.
Cycles 1- 4 are repeated up to 5 times using 5 different biotinylated antibodies.
(5) Imaging: The final slide is imaged using standard fluorescence microscopy, producing clear, high resolution multiplexed images for spatial analysis and biomarker discovery
