V5-tag and Nano-Trap

The V5-tag is a popular epitope tag derived from the P and V proteins of the Simian Virus 5 (SV5) that is used to tag proteins in yeast, bacteria, insect, and mammalian cells, allowing the detection and purification of the tagged protein. The V5-tag can be fused to the C-terminus or the N-terminus of the protein or be fused internally. Its amino acid sequence has a length of 14, GKPIPNPLLGLDST, with a molecular weight of 1.42 kDa.

Nano-Trap is ChromoTek's product line for the immunoprecipitation of tagged proteins. Their Nano-Trap line includes GFP-Trap, Myc-Trap, Spot-Trap, others, and since very recently, also V5-Trap. Nano-Trap differs from conventional affinity resin in the type of antibodies used: Nano-Trap antibodies are Nanobodies, very small antibodies derived from the heavy chain antibodies of camelids.

While anti-V5-tag antibodies for conventional IP are available from multiple suppliers, ChromoTek is the first to offer anti-V5 Nanobodies bound to agarose beads for immunoprecipitation. V5-Trap is available as agarose or magnetic agarose.
