Postdoc Webinar: Non-coding enhancer RNA as novel regulators of neuronal gene expression

Postdoc Webinar Series: LubioScience and GeneTex are honoured to host Dr. Nancy Carullo, a postdoctoral research fellow in the laboratory of the distinguished Prof. Dr. Isabelle Mansuy at the University of Zürich, Brain Research Institute. Dr. Carullo's work focuses on the characterization of activity-responsive eRNAs in neural cells and their role in gene regulation both, in cell culture and in the brain in the context of early-life stress (ELS).

The developing and adult mammalian brain is shaped by genetic and environmental factors and continuously adapts to external conditions throughout life. To function properly, the brain has to remain plastic while at the same time, store and use stable information. At the molecular level, this is achieved by adaptive transcriptional programs in brain cells. To coordinate these precise programs, the mammalian genome relies on gene regulatory elements known as enhancers. Enhancers are DNA sequences that regulate cell type- and tissue-specific gene expression, and provide exquisite spatial and temporal control of transcription. Active enhancers are pervasively transcribed to yield non-coding enhancer RNA (eRNA). A subgroup of enhancers and eRNA have recently been shown to respond to stimulation. eRNA possess unique properties to integrate information about external factors, interface with the coding however, whether eRNA contribute to transcriptional re-programing of brain cells after salient experiences is unknown. Dr. Carullo's work focuses on the characterizatoin of activity-responsive eRNAs in neural cells. She investigates their role in gene regulation both, in cell culture and in the brain the context of early-life stress (ELS). Her initial results suggest that the ELS alters expression of specific eRNA and that transcriptional manipulation of these eRNA with CRISPR tools mimics target gene expression in vitro.
