New FtsZ Proteins

 

 

FtsZ proteins are the bacterial homologues of tubulin and are essential for bacterial cell division.  FtsZ inactivation inhibits cell division, making them attractive targets for novel antimicrobial drugs.  Although FtsZ proteins exhibit a degree of homology, inhibitors of the proteins show differential affinity and differential efficacy. Thus, improved targeting can be achieved by screening several FtsZ proteins from different species. To improve drug screening of species-selective antimicrobials, Cytoskeleton is introducing FtsZ proteins from Staphylococcus aureus, Streptococcus pneumoniae and Enterococcus faecalis.

C-type Lectin Receptors CLRs
Multi-functional Dendritic Cell Markers and Regulators

 

C-type Lectin Receptors or CLRs are one of the major classes of Pattern Recognition Receptors (PRRs). CLRs are critical in recognition of antigens especially by Dendritic Cells (DCs) underscoring their importance as key regulatory immune receptors as well as markers for DCs1-2. Found in abundance especially on DCs as well as monocytes, macrophages and other myeloid cells, these receptors bind C-type lectins-conserved sugar structures on proteins or lipids from pathogens or self antigens alike. There are over 1,000 identified C-type lectin domain (CTLD) containing molecules to which CLRs bind in a Ca2+ dependent mechanism. The usual CTLD is composed of 2 separate loops connected by two disulfide bridges. The CLR binds to the CTLD usually on the 2nd loop3.

C-type lectins range in function as adhesion molecules, interactive molecules in the DC-T cell synapse, or in apoptosis. CLRs therefore serve as antigen receptors, regulators of cell trafficking and migration and controllers of DC-T cell immunoregulatory networks1-3.

As an example of a CLR functional marker on DCs, DC-SIGN binds HIV-1 and sequesters the virus in endosomes. HIV-1 laden DCs traffic to lymphoid tissues and interact with T cells affording their enhanced infection by the virus. DC-SIGN also binds CD50 (ICAM-3) allowing cell to cell contact in the DC-T cell synapse formation which drives immune responses3. Dectin-2, another CLR, binds mannose moieties and subsequently mediates DC cytokine production through NF-kB as well as through a SYK-CARD9 pathway4-5. Dectin-2 detection on monocytes is shown in the figure below.

Cell surface flow cytometric analysis of Dectin-2 expression by 5x10^5 human peripheral blood monocytes using 10 ul (0.25 ug) of IMG-6350D (anti-CD14), 10 ul (0.5 ug) of IMG-6682AF647 (anti-Dectin-2) and 0.5 ug of isotype control. IMGENEX’s cell surface flow kit (10084K) was used for this test.

IL-17 Measurements:
Which IL-17 Isoform or IL-17 Receptor is Important?

 

 

Ever since first being identified as a unique T cell subset, Th17 cells have been assigned a pivotal role in inflammatory diseases. As the hallmark cytokine produced by Th17 cells, IL-17 is an intensely studied inflammatory mediator used to identify and describe Th17 cell function. There are currently six proteins in the IL-17 family in mice and humans. Th17 cells produce and secrete IL-17 isoforms and pro-inflammatory cytokines in response to both exogenous and self antigens1-3 and there are five identified members of the IL-17 receptor (IL-17R) family, IL-17RA-IL-17RE, which bind multiple IL-17 proteins4. Most studied of the IL-17 protein family are the highly homologous IL-7A and IL-17F. Despite their homology to one another, binding to the same receptor, and overlap of similar functions, IL-17A and IL-17F are not interchangeable with one another. Several experimental models have demonstrated a predominance of IL-17A or IL-17F with minimal effect of the other. IL-17C and IL-17E are also known as other interleukins: IL-17C as IL-21 and IL-17E as IL-25. IL-25/IL-17E, is involved in allergic diseases and host defense against parasitic helminths and IL-17C is involved in pro-inflammatory activities, especially in autoimmune diseases5-7. So while IL-17A and IL-17F exert overlapping activities, it is important to distinguish the IL-17 isoforms and the receptors they bind to in order to delineate specific roles in overall host defense or autoimmune pathology.

Sensitivity: The Limit of Detection (LOD) [minimum detectable dose (MDD)] of IMGENEX‘s human IL-17A assay was determined by performing 20 replicates of assay diluent (blank) alone and calculating the mean signal + 2 standard deviations of the 20 values. The LOD is defined as the human IL-17A concentration corresponding to the mean assay blank + 2 SD. The LOD was determined to be < 1.95 pg/ml.

Spotlight on Dicer Knockout Embryonic Stem Cells

Dicer is an RNase III-containing enzyme that is central to the processing of most small RNAs.  Deletion of Dicer in mouse embryonic stem cells results in severe proliferation and differentiation defects.  A recent publication [PMID: 18923076] used Dicer knockout mouse embryonic stem cells as well as DGCR8 knockout mouse embryonic stem cells to studiy mammalian small RNA-generating pathways and shows that mammalian siRNAs exists in cell types other than oocytes. 

These Dicer and DGCR8 knockout cell lines are valuable research tools for scientists interested in studying the small non-coding RNAs in embryonic stem cells and the differentiated cells derived from them.

Description
Fibronectin purified from human plasma (lyophilized) 4 vials 5 mg each

Source

Human plasma. Donors tested negative for HBsAg, HCV, HIV1 and HIV2 antibodies


Purification

Affinity chromatography on gelatine agarose and heparin agarose


Format

Lyophilized from 4x2.5 ml of buffer (150 mM NaCl, 10 mM sodium phosphate pH 7.5). Non-sterile.


Reconstitution

Add 4x5 ml of warm (37°C) PBS . Incubate at 37°C for 3 hours, occasionally inverting the vial. Do not vortex. Reconstituted solution contains 4x5 mg of soluble fibronectin and a minor quantity of insoluble fibronectin aggregate. Filter slowly through 0.2 µm membrane filter. Concentration 1 mg/ml of fibronectin corresponds to A280 - 1.3

Spotlight on LDL and VLDL Receptors

Low density lipoprotein (LDL) and very low density lipoprotein (VLDL) receptors are cell-surface receptor proteins that regulate endocytosis of cholesterol and lipoproteins into mammalian cells. These receptors are essential for proper lipid metabolism and healthy cell function. However, LDLR and VLDLR are also associated with a wide variety of diseases, due to their role as gateways between external plasma and cells.
A 2011 study in Hepatology analyzed the role of the LDLR in the Hepatitis C virus life cycle, and found that the physiological function of LDLR is necessary for optimal replication of the Hepatitis C virus genome. Mutations in the LDLR gene cause the autosomal dominant disorder,hypercholesterolemia. Alternatively, defects in VLDLR can cause cerebellar ataxia, mental retardation and dysequilibrium syndrome type 1. VLDLR may also be a susceptibility gene for Alzheimer diseases due to the Reelin binding that induces modulation of Tau phosphorylation.
