Purification

Affinity chromatography on gelatine agarose and heparin agarose


Format

Lyophilized from 4x2.5 ml of buffer (150 mM NaCl, 10 mM sodium phosphate pH 7.5). Non-sterile.


Reconstitution

Add 4x5 ml of warm (37°C) PBS . Incubate at 37°C for 3 hours, occasionally inverting the vial. Do not vortex. Reconstituted solution contains 4x5 mg of soluble fibronectin and a minor quantity of insoluble fibronectin aggregate. Filter slowly through 0.2 µm membrane filter. Concentration 1 mg/ml of fibronectin corresponds to A280 - 1.3

Spotlight on LDL and VLDL Receptors

Low density lipoprotein (LDL) and very low density lipoprotein (VLDL) receptors are cell-surface receptor proteins that regulate endocytosis of cholesterol and lipoproteins into mammalian cells. These receptors are essential for proper lipid metabolism and healthy cell function. However, LDLR and VLDLR are also associated with a wide variety of diseases, due to their role as gateways between external plasma and cells.
A 2011 study in Hepatology analyzed the role of the LDLR in the Hepatitis C virus life cycle, and found that the physiological function of LDLR is necessary for optimal replication of the Hepatitis C virus genome. Mutations in the LDLR gene cause the autosomal dominant disorder,hypercholesterolemia. Alternatively, defects in VLDLR can cause cerebellar ataxia, mental retardation and dysequilibrium syndrome type 1. VLDLR may also be a susceptibility gene for Alzheimer diseases due to the Reelin binding that induces modulation of Tau phosphorylation.

Spotlight on CASP8 and STAG2

Malignant melanoma, the most dangerous type of skin cancer, is estimated to affect roughly one in every 65 adults. Although it is typically preventable, the only treatment is surgical tumor excision. Unfortunately, it is often difficult to prevent the cancer from spreading due to melanoma's high malignancy rates.

Scientists at the University of Leeds recently found that a DNA defect in the apoptosis mediator gene CASP8 significantly increases the risk of melanoma [PMID: 21983787]. Additionally, researchers at the Georgetown University School of Medicine discovered that one fifth of melanomas have a defective copy of the STAG2 Gene, which plays a role in the division of genetic material between cells. [PMID: 2021852505]. These exciting new discoveries offer promise for potential future treatments, and open important new avenues for melanoma research.

Spotlight on Beclin 1

Beclin 1 is a 60kDa, BH3-only domain autophagy protein that is essential for the regulation of macroautophagy and the formation of the autophagic vacuole. Through a diverse network of protein interactions, Beclin 1 appears to be a key player in the regulation of a number of critical cellular processes. Specifically, it is a key component of the class III PI3K complex, and interacts with the anti-apoptotic protein, BcL2. Beclin 1 contains 3 identical structural domains comprised of an N-terminal BH3 domain, a central coiled-coil domain and an evolutionarily conserved domain. The expression of Beclin 1 appears to be generally ubiquitous in human and mouse tissues and it is localized predominantly within cytoplasmic structures such as the mitochondria, ER and the perinuclear membrane. Novus offers more than 30 Beclin 1 antibodies for a wide variety of species and applications, which serve as useful tools for autophagy research.

Oxford Biomedical Research has recently redesigned its FR40 TBARS assay to increase sensitivity and reduce interferences.  We have also introduced a fluorometric TBARS assay with exceptional sensitivity.  These kits have the best sensitivity and the least interference of all kits commercially available.  While all TBARS assays detect endogenous interfering factors, many more are generated when samples are boiled with strong acid.  Their new, patent pending method uses a milder acid and can be run at room temperature.

Novus Biologicals announced today a new XBP1 Antibody (NBP1-77681) for use in immune response and endoplasmic reticulum stress research.

X-box binding protein 1 (XBP1) is a nuclear transcription factor which activates the unfolded protein response during endoplasmic reticulum stress response. Specifically, XBP-1 is upregulated during ER stress, and directly binds the unfolded protein response element to induce mis-folding of several downstream proteins.

XBP-1 is also essential for the growth and differentiation of hepatocyte and plasma cells, as well as some brain development and cognitive ability formation. It has been implicated in a wide range of disorders, including Alzheimer’s disease, major affective disorder type 7, Crohn’s disease and IBS.

Spotlight on Jumonji

Jumonji, Japanese for cruciform, was originally named for the appearance of the neural plates in jumonji mutant nice, and means cruciform in Japanese. Within the protein, jumonji contains an AT-rich interacting domain, a jmjN domain, and a jmjC domain. Since the discovery of this gene, jmjC domains have been found in over 100 other proteins, and are evolutionarily conserved from human to yeast. JmjC domains play a role in stem cell differentiation and renewal, tumor suppression, obesity resistance, and gene expression.

Recently, some jumonji family members have been identified as histone demethylases. Jumonji member JMJD1A specifically demethylates mono and dimethyl forms of histone 3 Lysine 9. Another member, JMJD2A also works specifically on H3 Lys9, but in the trimethylated form, along with trimethyl H3 Lys36. JMJD2C also works on these sites, but is specific for the trimethyl forms.

New TP53INP2 & GABARAP Antibodies

Novus Biologicals announced new antibodies to TP53INP2 (NBP1-71704) and GABARAP
(NBP1-71771) for use in autophagy research.

Tumor Protein 53-Induced Nuclear Protein 2 (TP53INP2) is thought to be a scaffold protein which translocates from the nucleus to autophagosomes during preparation for autophagy. TP53INP2 binds LC3-related proteins, including GABARAP and VMP1. Research has shown that both TP53INP2 and GABARAP are required for autophagy in mammalian cells.

Spotlight on Pancreatic Cancer Stem Cells

A recent study done at the Spanish National Cancer Research Center (CNIO), published in the November issue of Cell Stem Cell [PMID:22056140] , successfully demonstrates a therapeutic strategy for targeting pancreatic cancer stem cells (CSC).  Their research shows that the Nodal/Activin pathway is critical for the self-renewal capacity and the stemness properties of pancreatic CSC.  In addition, Nodal/Activin is strongly expressed by pancreatic CSC, and also by pancreatic stellate cells (PSC).  PSC are abundant in the stroma that surrounds the pancreatic CSC, and may potentially be the supportive niche for these CSC.  They demonstrated that by targeting the stroma as well as the CSC they were able to effectively treat pancreatic cancer in a number of preclinical models.

Spotlight on Heat Shock Protein 70

Excessive environmental stress induces a complex cellular reaction known as the heat shock response, to protect proteins from damage. A special class of proteins called Heat Shock Proteins (HSP) is significantly upregulated upon cellular stress, such as hypoxia, infection, inflammation, or starvation. Of the three most widely studied HSPs (HSP60, HSP70 & HSP90), HSP70 has begun to emerge as critical factor in the hypoxic stress response mechanism.

Many researchers have shown that HSP70 expression is strongly increased in cells exposed to hypoxic conditions. However, recent studies have revealed more about the functional role that HSP70 plays in the heat shock response and the protective effects that it confers on the cell.
